Tirzepatide is a synthetic peptide built from thirty-nine amino acids. Its sequence is derived from native glucose-dependent insulinotropic polypeptide, or GIP, with several non-natural residues and a fatty diacid side chain attached through a linker. The molecule behaves as a dual agonist at two incretin receptors, GIP and GLP-1, instead of targeting a single receptor. This dual engagement separates it from earlier single-receptor incretin compounds and underpins most of its reported pharmacological activity.
At the receptor level, the compound binds both GIP and GLP-1 receptors and triggers downstream signalling that raises cyclic AMP in target cells. GLP-1 receptor activation is associated with glucose-dependent insulin release, slower gastric emptying, and reduced appetite signalling. GIP receptor activation contributes effects that are less completely characterised, and how much each receptor adds to the overall clinical response is still an open question. The two pathways appear to interact in a complementary rather than a purely additive way.
The peptide shares degradation routes common to modified peptides: deamidation of asparagine and glutamine residues, oxidation of methionine, and backbone hydrolysis under extreme pH. Lyophilized material is generally more stable than a solution, and residual water content directly affects the rate of hydrolysis. In liquid form, aggregation and visible particles can appear after agitation or repeated freeze-thaw cycles. Stability studies therefore track monomer content, aggregate content, and potency over months under defined temperature and humidity.
Cold-chain handling is standard for formulated product, with dry powder stored frozen and ready-to-use solutions refrigerated. Light exposure is minimized because photodegradation of certain amino acid side chains is possible. Shipping and temperature-excursion studies are used to establish whether short deviations affect quality attributes. Documentation supplied with research material usually includes a certificate of analysis listing purity, identity confirmation, and water or residual solvent content. Users are expected to confirm that material meets the stated specification before use.

Verification of research-grade material involves checking purity, sequence and counter-ion content against a certificate of analysis. Reported purity figures usually reflect chromatographic area percentage and do not by themselves establish biological activity. Independent laboratories may repeat mass confirmation and peptide mapping to detect substitutions or truncations. Open questions concern how residual solvents, trace metals and subtle conformational variants affect measured behavior, and how consistently different suppliers define their specifications. Documentation of analytical methods matters as much as the headline purity number when results are compared across studies.
Routine characterization relies on reversed-phase high-performance liquid chromatography, often coupled to mass spectrometry, to confirm identity and estimate purity. Peptide mapping after enzymatic digestion verifies the amino acid sequence and locates appended groups such as the fatty acid chain. Size-exclusion chromatography detects aggregates and fragments, while ion-exchange chromatography resolves charge variants. Circular dichroism and nuclear magnetic resonance supply secondary and higher-order structural information in research settings. No single technique covers every attribute, so laboratories combine orthogonal methods and compare outcomes against a reference standard where one exists.
Purified material is typically handled as a lyophilized powder kept at or below minus twenty degrees Celsius, shielded from light and moisture. In that state the solid remains stable for extended periods, although repeated freeze-thaw cycling can encourage aggregation. Once dissolved, aqueous solutions are less durable and are generally held cold and used within a brief window. Buffer composition, pH and ionic strength all influence degradation rates, and mildly acidic to neutral conditions are commonly examined. Actual shelf life depends on formulation, concentration and container, so stability limits are established experimentally rather than assumed.
Receptor activation by tirzepatide raises intracellular cyclic AMP through Gs-coupled signalling at both targets. At the GLP-1 receptor the downstream effect includes glucose-dependent insulin release, suppressed glucagon secretion, delayed gastric emptying, and reduced appetite signalling in the hypothalamus. GIP receptor engagement adds insulinotropic activity and appears to influence lipid handling in adipose tissue. Because both receptors are stimulated at the same time, the pharmacological profile differs from that of selective GLP-1 receptor agonists, and the relative contribution of each arm remains an area of active investigation.
Clinical development proceeded through large phase 3 programmes in type 2 diabetes and in obesity or overweight with at least one weight-related comorbidity. Regulatory approvals followed in several jurisdictions for both indications. Weekly subcutaneous dosing reflects an elimination half-life of roughly five days. Open questions include the durability of metabolic effects after treatment stops, long-term cardiovascular and hepatic outcomes beyond completed trials, and whether the dual mechanism confers benefits independent of total receptor occupancy. Published literature continues to expand on these points. Substantial uncertainty remains about interindividual variability in response.
Radar equipment is used at airports, in airplanes, at missile sites, on tanks, and on ships. The radar technology commonly used in the 20th century produced X-rays as a technically unavoidable by-product in the high-voltage electronics of the equipment. In the 1960s and 1970s, German soldiers and technicians were largely unaware of the dangers, as were those in the GDR's National People's Army. The problem had been known internationally since the 1950s, and to the German Armed Forces since at least 1958. However, no radiation protection measures were taken, such as the wearing of lead aprons. Until about the mid-1980s, radiation shielding was inadequate, especially for pulse switch tubes. Particularly affected were maintenance technicians (radar mechanics) who were exposed to the X-ray generating parts for hours without any protection. The permissible annual limit value could be exceeded after just 3 minutes. It was not until 1976 that warning notices were put up and protective measures taken in the German Navy, and not until the early 1980s in general. As late as the 1990s, the German Armed Forces denied any connection between radar equipment and cancer or genetic damage. The number of victims amounted to several thousand. The connection was later acknowledged by the German Armed Forces and in many cases a supplementary pension was paid. In 2012, a foundation was set up to provide unbureaucratic compensation for the victims.
Rudolf Aebersold (better known as Ruedi Aebersold; born 12 September 1954) is a Swiss biologist, regarded as a pioneer in the fields of proteomics and systems biology. He has primarily researched techniques for measuring proteins in complex samples, in many cases via mass spectrometry. Ruedi Aebersold is a professor of Systems biology at the Institute of Molecular Systems Biology (IMSB) in ETH Zurich. He was one of the founders of the Institute for Systems Biology in Seattle, Washington, United States where he previously had a research group. Aebersold is known for the development and application of targeted proteomics techniques in the field of biomedical research, in order to understand the function, interaction and localization of each protein in the cell and its changes in disease states. To this end, Ruedi Abersold has made significant contributions in the development and application of targeted proteomics methods, including selected reaction monitoring and data-independent acquisition. Ruedi Aebersold is also recognized for its contributions in the development of standard formats and open source software for the analysis and storage of mass spectrometry and proteomics data, and he is one of the inventors of the Isotope-Coded Affinity Tag (ICAT) technique for quantitative proteomics, a technique that measures the relative quantities of proteins between two sample by using tags containing stable isotopes of different masses. Aebersold is co-founder and scientific advisor of the companies ProteoMediX and Biognosys.
Microtubule-disruptive drugs like vinblastine, colcemid, and nocodazole have been reported to act by two mechanisms. At very low concentrations they suppress microtubule dynamics and at higher concentrations they reduce microtubule polymer mass. Recent findings indicate that they also produce microtubule fragments by stimulating microtubule minus-end detachment from their organizing centers. Dose-response studies further indicate that enhanced microtubule detachment from spindle poles correlate best with cytotoxicity. But research into the mechanism is still ongoing as recent studies also show vinblastine inducing apoptosis that is phase-independent in certain leukemias.
=== 2014—present: Australian opening === On 31 March 2014, Blumenthal announced he would close the Fat Duck for renovations for six months and temporarily relocate it with its entire team to Crown Towers in Melbourne, Australia. During those six months, the Australian restaurant was also named the Fat Duck, after which it was renamed after Blumenthal's London restaurant Dinner. It was the second restaurant with that name, Blumenthal's sixth restaurant and his first restaurant outside of Britain. This temporary closure of the Bray location made the Fat Duck ineligible for assessment for the 2016 Michelin Guide, thus losing its three-starred status. It regained the stars the following year. As of 2025, Blumenthal was spending more time at the Fat Duck than he had done over the preceding 20 years, and was preparing a menu for its 30th anniversary.

The United States investigated ricin for its military potential during World War I. At that time it was being considered for use either as a toxic dust or as a coating for bullets and shrapnel. The dust cloud concept could not be adequately developed, and the coated bullet/shrapnel concept would violate the Hague Convention of 1899 (adopted in U.S. law at 32 Stat. 1903), specifically Annex §2, Ch.1, Article 23, stating "... it is especially prohibited ... [t]o employ poison or poisoned arms". In 'Ricin and Abrin', Manashi Bagchi, Shirley Zafra-Stone, Francis C. Lau, and Debasis Bagchi wrote that during World War II the United States and Canada studied ricin in cluster bombs. Though there were plans for mass production and several field trials with different bomblet concepts, the end conclusion was that it was no more economical than using phosgene. This conclusion was based on comparison of the final weapons, rather than ricin's toxicity (LCt50 ~10 mg/min·m3). Ricin was given the military symbol W or later WA. Interest in it continued for a short period after World War II, but soon subsided when the US Army Chemical Corps began a program to weaponize sarin. The Soviet Union possessed weaponized ricin. The KGB developed weapons using ricin which were used outside the Soviet bloc, most famously in the Markov assassination.
DBNPA dust may cause mechanical (abrasive) irritation to the skin, eyes and respiratory system and is severely irritating to the eyes and may cause permanent corneal damage. Skin contact may cause an allergic skin reaction. Pre-existing skin disorders may even be aggravated by over-exposure to this product. Regarding long-term health effects, DBNPA contains material that may cause target organ damage, based on animal data. Repeated or prolonged inhalation of DBPNA dust can lead to chronic respiratory problems. Once sensitized, an allergic skin reaction may occur when subsequently exposed to very low levels of DBNPA.
Soon after inoculation, yeast begin to rapidly consume the available assimilable nitrogen with up to 46% of YAN being fully consumed by the onset of full fermentation. Because inorganic nitrogen, such as the ammonium salts in DAP, are toxic to yeast in high levels, it is never added during inoculation when the biomass of the newly re-hydrated yeast is low. Many winemakers split up the dosage of DAP with the first addition being made at the end of the lag phase when the yeast enter their period of exponential growth and alcoholic fermentation begins. In most must this is around 48 to 72 hours after inoculation. A second dosage is then often added around a third of the way through sugar fermentation and often before the sugar levels hit 12-10 Brix (6.5 to 5.5 Baumé, 48.3 to 40.0 Oechsle) because as the fermentation progresses yeast cells are no longer able to bring the nitrogen into the cell due to the increasing toxicity of ethanol surrounding the cells. This leaves the nitrogen unused and available for spoilage organisms that may come afterwards.
At community gatherings, target shooting was a major sport; they practised shooting at targets, such as hens' eggs perched on posts 100 metres (110 yd) away. They made expert mounted infantry, using cover, from which they could pour in a destructive fire using modern, smokeless, Mauser rifles. In preparation for hostilities, the Boers had acquired around 100 of the latest Krupp field guns, all horse-drawn and dispersed among the Kommando groups and several Le Creusot "Long Tom" siege guns. The Boers' skill in adapting themselves to become first-rate artillerymen shows that they were a versatile adversary. The Transvaal had an intelligence service that stretched across South Africa, and of whose extent and efficiency the British were as yet unaware.
== Screening == Initial screening for CIP/CIM may be performed using an objective scoring system for muscle strength. The Medical Research Council (MRC) score is one such tool, and sometimes used to help identify CIP/CIM patients in research studies. The MRC score involves assessing strength in 3 muscle groups in the right and left sides of both the upper and lower extremities. Each muscle tested is given a score of 0–5, giving a total possible score of 60. An MRC score less than 48 is suggestive of CIP/CIM. However, the tool requires that patients be awake and cooperative, which is often not the case. Also, the screening tool is non-specific, because it does not identify the cause a person's muscle weakness. Once weakness is detected, the evaluation of muscle strength should be repeated several times. If the weakness persists, then a muscle biopsy, a nerve conduction study (electrophysiological studies), or both should be performed.

It acts as a dual agonist at the GIP receptor and the GLP-1 receptor. This broader targeting profile distinguishes it from selective GLP-1 agonists, which engage only one receptor.
A fatty diacid side chain promotes binding to albumin, which delays clearance from circulation. The half-life of roughly five days makes a weekly schedule practical.
No. It is a synthetic peptide whose backbone is based on the natural incretin hormone GIP. Non-natural residues and the lipid side chain were engineered to improve stability and duration of action.
Reversed-phase liquid chromatography with ultraviolet detection is the usual approach, frequently combined with mass spectrometry for identity. Purity is reported as the area percentage of the main peak. Related impurities eluting near the main peak are usually summed and reported separately.
