This is a working overview of deamidation, written for readers who want more than a one-paragraph summary but less than a textbook.
This page was last updated on 2026-07-03 and is reviewed periodically as new material appears.
Identity and purity are assessed by reversed-phase high-performance liquid chromatography, with mass confirmation by electrospray ionisation mass spectrometry. Peptide mapping after enzymatic digestion verifies the primary sequence. Size-exclusion chromatography quantifies aggregates, while circular dichroism provides a secondary-structure fingerprint. Bioanalytical quantification in plasma uses immunoassay or LC-MS/MS. Reported purity for research-grade lots is commonly 95 percent or higher, and residual water content is checked by Karl Fischer titration.
As a peptide, tirzepatide is handled as a lyophilised solid in research settings and as a preserved solution in finished products. Aqueous solubility is pH dependent and reaches a minimum near the isoelectric point, which lies close to pH 5.4. Stock solutions are typically prepared in neutral or slightly basic buffer to limit precipitation. The solid is hygroscopic and should be equilibrated to room temperature before opening so that condensation does not form on the powder surface.
Recommended storage for reference material is a freezer at approximately -20 degrees Celsius, protected from light and moisture. Commercial injectable presentations are stored refrigerated between 2 and 8 degrees Celsius and must not be frozen. Product labelling generally permits a limited period at controlled room temperature once dispensed, with the exact window depending on the presentation. Repeated temperature cycling is avoided because it can promote aggregation or deamidation of the peptide chain.

Purified material is typically handled as a lyophilized powder kept at or below minus twenty degrees Celsius, shielded from light and moisture. In that state the solid remains stable for extended periods, although repeated freeze-thaw cycling can encourage aggregation. Once dissolved, aqueous solutions are less durable and are generally held cold and used within a brief window. Buffer composition, pH and ionic strength all influence degradation rates, and mildly acidic to neutral conditions are commonly examined. Actual shelf life depends on formulation, concentration and container, so stability limits are established experimentally rather than assumed.
Verification of research-grade material involves checking purity, sequence and counter-ion content against a certificate of analysis. Reported purity figures usually reflect chromatographic area percentage and do not by themselves establish biological activity. Independent laboratories may repeat mass confirmation and peptide mapping to detect substitutions or truncations. Open questions concern how residual solvents, trace metals and subtle conformational variants affect measured behavior, and how consistently different suppliers define their specifications. Documentation of analytical methods matters as much as the headline purity number when results are compared across studies.
Routine characterization relies on reversed-phase high-performance liquid chromatography, often coupled to mass spectrometry, to confirm identity and estimate purity. Peptide mapping after enzymatic digestion verifies the amino acid sequence and locates appended groups such as the fatty acid chain. Size-exclusion chromatography detects aggregates and fragments, while ion-exchange chromatography resolves charge variants. Circular dichroism and nuclear magnetic resonance supply secondary and higher-order structural information in research settings. No single technique covers every attribute, so laboratories combine orthogonal methods and compare outcomes against a reference standard where one exists.
Lyophilized material is generally held at -20 degrees Celsius or lower, desiccated and protected from light, where it remains stable for extended periods. Reconstituted or ready-to-use solution is usually kept at 2 to 8 degrees Celsius with minimal agitation. Repeated freeze-thaw cycles should be avoided because they promote aggregation and reduce the soluble monomer fraction. Shipment of frozen solid commonly uses dry ice, while refrigerated liquid moves with validated cold packs. Stability beyond documented periods is not established.
Characterization of the peptide relies on reversed-phase high-performance liquid chromatography for purity and related-substance profiling, with ultraviolet detection near 214 nanometers. Mass spectrometry confirms molecular mass and reveals modifications such as oxidation or deamidation. Peptide mapping after enzymatic digestion verifies the amino acid sequence, while amino acid analysis supplies compositional data. Circular dichroism and infrared spectroscopy are used to assess secondary structure, particularly the alpha-helical content that influences aggregation behavior in solution.

The university's mascots are a pair of anthropomorphized wildcats named Wilbur and Wilma. The human figures behind Wilbur and Wilma are kept secret through the year as the mascots appear only in costume. In 1986, Wilbur married his longtime wildcat girlfriend, Wilma. Together, Wilbur and Wilma appear along with the cheerleading squad at most Wildcat sporting events. Arizona's first mascot was a real desert bobcat named "Rufus Arizona", introduced in 1915.

Cell phone – a portable wireless telephone that is connected to the telephone network by radio signals exchanged with a local antenna at a cellular base station (cell tower). The service area covered by the provider is divided into small geographical areas called "cells", each served by a separate base station antenna and multichannel transceiver. All the cell phones in a cell communicate with this antenna on separate frequency channels, assigned from a common pool of frequencies. The purpose of cellular organization is to conserve radio bandwidth by frequency reuse. Low power transmitters are used so the radio waves used in a cell do not travel far beyond the cell, allowing the same frequencies to be reused in geographically separated cells. When a user carrying a cellphone crosses from one cell to another, his phone is automatically "handed off" seamlessly to the new antenna and assigned new frequencies. Cellphones have a highly automated full duplex digital transceiver using OFDM modulation using two digital radio channels, each carrying one direction of the bidirectional conversation, as well as a control channel that handles dialing calls and "handing off" the phone to another cell tower. Older 2G, 3G, and 4G networks use frequencies in the UHF and low microwave range, between 700 MHz and 3 GHz. The cell phone transmitter adjusts its power output to use the minimum power necessary to communicate with the cell tower; 0.6 W when near the tower, up to 3 W when farther away. Cell tower channel transmitter power is 50 W.

primer A short, single-stranded oligonucleotide, typically 5–100 bases in length, which "primes" or initiates nucleic acid synthesis by hybridizing to a complementary sequence on a template strand and thereby providing an existing 3'-end from which a polymerase can extend the new strand. Natural systems exclusively use RNA primers to initiate DNA replication and some forms of prokaryotic transcription, whereas the in vitro syntheses performed in many laboratory techniques such as PCR often use DNA primers. In modern laboratories, primers are carefully designed, often in "forward" and "reverse" pairs, to complement specific and unique sequences in target DNA molecules, with consideration given to their melting and annealing temperatures, and then purchased from commercial suppliers which create oligonucleotides on demand by de novo synthesis.
In many MHD systems, most of the electric current is compressed into thin, nearly two-dimensional ribbons termed current sheets. These can divide the fluid into magnetic domains within which the currents are relatively weak. Current sheets in the solar corona are thought to be between a few meters and a few kilometers thick, which is quite thin compared to the magnetic domains, which are thousands to hundreds of thousands of kilometers across. Another example occurs in the Earth's magnetosphere, where current sheets separate topologically distinct domains, isolating most of the Earth's ionosphere from the solar wind.

== Life == Sun Yaoting was born in a village near Tianjin, about 60 miles southeast of Beijing. Born into poverty, his father was a jianbing peddler. When he was young, his family's home was burned down and their fields were stolen by a landlord in their village. Inspired by an older eunuch from his village who had become rich, coupled with the family's hope that they would gain Imperial influence and get revenge on the landlord, the family decided to emasculate Sun. In 1911, at the age of eight, Sun was emasculated by his father at home on their bed with a single razor cut, without the use of anesthesia. A piece of oil-soaked paper was used as a bandage, while a goose quill was inserted in Sun's urethra to prevent it from getting blocked as the wound healed. Sun remained unconscious for three days and could barely move for two months. However, the emperor he had hoped to serve, Puyi, had abdicated several weeks before his recovery during the Xinhai Revolution. Nonetheless, Sun went to Beijing at age 14. His rank initially was so low that he was not even given a name, and worked as a servant to another, higher ranking eunuch. He then rose through the ranks, becoming a palace eunuch serving under one of the emperor’s uncles, eventually becoming an attendant to the Qing empress. Corruption amongs the eunuchs working in the Forbidden City was rampant, as they constantly stole treasures and sold it on the black market.

=== Macromolecular therapeutics for brain delivery === Nanomedicines designed for brain delivery/action have a difficult hurdle to overcome; they need to cross the blood brain barrier. We focus on receptor binding peptides that transcytose bound cargo into the brain. In particular, angiopep-2 (TFFYGGSRGKRNNFKTEEY) binds to LDLR (low-density lipoprotein receptor)-related protein (LRP)-1 followed by transcytosis. In collaboration with the University of Utah Department of Radiology we are developing conjugates suitable for the treatment of traumatic brain injury and Alzheimer disease.
It is thought that high glucagon levels and lack of insulin production are the main triggers for the metabolic issues associated with Type I diabetes, in particular maintaining normal blood glucose levels, formation of ketone bodies, and formation of urea. One finding of note is that the glucagon response to hypoglycemia is completely absent in patients with Type I diabetes. Consistently high glucagon concentrations in the blood can lead to diabetic ketoacidosis, which is when ketones from lipid breakdown build up in the blood, which can lead to dangerously low blood glucose levels, low potassium levels, and in extreme cases cerebral edema. It has been proposed that the reason for the high levels of glucagon found in the plasma of patients with Type I diabetes is the absence of beta cells producing insulin and the reciprocal effect this has on delta cells and the secretion of somatostatin.
==== In aqueous solution ==== Most neptunium coordination complexes known in solution involve the element in the +4, +5, and +6 oxidation states: only a few studies have been done on neptunium(III) and (VII) coordination complexes. For the former, NpX2+ and NpX+2 (X = Cl, Br) were obtained in 1966 in concentrated LiCl and LiBr solutions, respectively: for the latter, 1970 experiments discovered that the NpO3+2 ion could form sulfate complexes in acidic solutions, such as NpO2SO+4 and NpO2(SO4)−2; these were found to have higher stability constants than the neptunyl ion (NpO2+2). A great many complexes for the other neptunium oxidation states are known: the inorganic ligands involved are the halides, iodate, azide, nitride, nitrate, thiocyanate, sulfate, carbonate, chromate, and phosphate. Many organic ligands are known to be able to be used in neptunium coordination complexes: they include acetate, propionate, glycolate, lactate, oxalate, malonate, phthalate, mellitate, and citrate. Analogously to its neighbours, uranium and plutonium, the order of the neptunium ions in terms of complex formation ability is Np4+ > NpO2+2 ≥ Np3+ > NpO+2. (The relative order of the middle two neptunium ions depends on the ligands and solvents used.) The stability sequence for Np(IV), Np(V), and Np(VI) complexes with monovalent inorganic ligands is F− > H2PO−4 > SCN− > NO−3 > Cl− > ClO−4; the order for divalent inorganic ligands is CO2−3 > HPO2−4 > SO2−4. These follow the strengths of the corresponding acids. The divalent ligands are more strongly complexing than the monovalent ones.

Solid material is normally kept frozen at about -20 degrees Celsius, desiccated and protected from light. Solutions are held cold and used within a defined window because degradation products accumulate over time.
Mass spectrometry, usually electrospray ionisation coupled to liquid chromatography, gives an observed mass for comparison with the theoretical value. Peptide mapping after digestion provides complementary sequence-level confirmation.
Short exposure during weighing and handling is generally tolerated, but prolonged storage above refrigerated conditions raises the risk of deamidation, oxidation, and aggregation. The rate depends on pH, buffer composition, and concentration.
建议在低温、避光、干燥环境中密封保存,常见条件为 2 至 8 摄氏度,长期存放可置于更低温度。应避免反复冻融,并尽量减少容器开启次数。
