== Covalently modulated enzymes == Here, the active and inactive form of the enzymes are altered due to covalent modification of their structures which is catalysed by other enzymes. This type of regulation consists of the addition or elimination of some molecules which can be attached to the enzyme protein. The most important groups that work as modifiers are phosphate, methyl, uridine, adenine and adenosine diphosphate ribosyl. These groups are joined to or eliminated from the protein by other enzymes. The most remarkable covalent modification is phosphorylation. Serine, Threonine and Tyrosine are common amino acids that participate in covalent modifications and are used to control enzyme’s catalytic activities. Kinase and phosphatases are commonly known enzymes that affect these modifications, which result in shifting of conformational states of the binding affinity to substrate.

== Receptor == The endogenous receptor for Substance P is neurokinin 1 receptor (NK1-receptor, NK1R). It belongs to the tachykinin receptor sub-family of GPCRs. Other neurokinin subtypes and neurokinin receptors that interact with SP have been reported as well. Amino acid residues that are responsible for the binding of SP and its antagonists are present in the extracellular loops and transmembrane regions of NK-1. Binding of SP to NK-1R results in internalization by the clathrin-dependent mechanism to the acidified endosomes where the complex disassociates. Subsequently, SP is degraded and NK-1R is re-expressed on the cell surface. Substance P and the NK1-receptor are widely distributed in the brain and are found in brain regions that are specific to regulating emotion (hypothalamus, amygdala, and the periaqueductal gray). They are found in close association with serotonin (5-HT) and neurons containing norepinephrine that are targeted by the currently used antidepressant drugs. Argentina took "speedy coffee" before the qualifier for the 1994 World Cup against Australia, at least this is what Maradona said in May 2011. It should make them run faster, but also caused sleeping problems. He also found it suspicious that only the deciding match (against Australia) had no anti-doping control. Grondona, chairman of AFA back then, responded that there were no tests because Maradona, who already had a drug history, might not have passed. Maradona tested positive in the world cup. Immediately after the World Cup 1998, all of the drug testing samples were destroyed. If the same had happened in the Tour de France, former WADA director Alain Garnier argued that Lance Armstrong would not have been caught. Marie-George Buffet, sports minister at that time, also recalls that she felt pressurised when she initiated an unannounced test in December 1997. There were no more unannounced tests after that. Jean-Pierre Paclet, physician of Les Bleus in 1998, mentions "abnormal haematocrit values" in his book. == Amino acid propensities == The original Chou–Fasman parameters found some strong tendencies among individual amino acids to prefer one type of secondary structure over others. Alanine, glutamate, leucine, and methionine were identified as helix formers, while proline and glycine, due to the unique conformational properties of their peptide bonds, commonly end a helix. The original Chou–Fasman parameters were derived from a very small and non-representative sample of protein structures due to the small number of such structures that were known at the time of their original work. These original parameters have since been shown to be unreliable and have been updated from a current dataset, along with modifications to the initial algorithm. The Chou–Fasman method takes into account only the probability that each individual amino acid will appear in a helix, strand, or turn. Unlike the more complex GOR method, it does not reflect the conditional probabilities of an amino acid to form a particular secondary structure given that its neighbors already possess that structure. This lack of cooperativity increases its computational efficiency but decreases its accuracy, since the propensities of individual amino acids are often not strong enough to render a definitive prediction.
