In clinical research, tirzepatide has been studied in randomized controlled trials for glycemic control and body weight reduction. These trials typically measure changes in hemoglobin A1c and body weight over periods of several months. The drug is administered by subcutaneous injection, and its pharmacokinetic profile supports once-weekly dosing. Post-marketing surveillance continues to evaluate long-term outcomes and rare adverse events.
Tirzepatide is a synthetic peptide that acts as a dual agonist at the glucose-dependent insulinotropic polypeptide (GIP) and glucagon-like peptide-1 (GLP-1) receptors. The molecule contains 39 amino acids and features a C20 fatty diacid moiety attached via a linker, which promotes albumin binding and extends its circulating half-life. Its sequence incorporates non-natural amino acids and modifications that reduce susceptibility to degradation by dipeptidyl peptidase-4. This dual receptor activity distinguishes it from selective GLP-1 receptor agonists.

The peptide activates two G protein-coupled receptors, GIPR and GLP-1R. Binding triggers adenylyl cyclase activity and raises intracellular cyclic AMP in pancreatic beta cells, which potentiates insulin release when glucose is elevated. Signaling in the central nervous system is associated with reduced appetite and lower energy intake, while effects on gastric emptying and glucagon secretion are also reported. Because activity at both receptors is retained, the pharmacological profile is often described as incretin-based rather than selective for a single receptor.
After subcutaneous injection, absorption is gradual, and peak plasma levels are generally reached within one to three days. Albumin binding extends the apparent half-life to roughly five days, which supports a weekly administration schedule. Metabolism proceeds mainly through proteolytic cleavage of the peptide backbone and beta-oxidation of the fatty acid chain, rather than through cytochrome P450 pathways. Eliminated fragments are largely recycled through general protein turnover, and excretion of intact drug in urine is minimal. These properties distinguish the molecule from short-acting incretin mimetics.
Routine characterization of the peptide relies on reversed-phase high-performance liquid chromatography for purity assessment, usually with ultraviolet detection near 214 nanometers. Intact mass measurement by liquid chromatography coupled to mass spectrometry confirms molecular identity against a theoretical value. Sequence-level confirmation uses enzymatic digestion followed by tandem mass spectrometry, an approach known as peptide mapping. Amino acid analysis gives an independent check on composition. Circular dichroism spectra are used to estimate helical content in aqueous buffer.
Stability depends strongly on physical form. The dry powder is generally regarded as stable for extended periods when held at or below minus twenty degrees Celsius in a sealed, desiccated container. In solution, degradation pathways include deamidation of asparagine and glutamine residues, oxidation of methionine, and aggregation. Reaction rates for these pathways rise with temperature. Repeated freezing and thawing of solutions promotes aggregation, and light exposure can accelerate some oxidative changes. Buffer composition and pH influence which pathway dominates at a given temperature.
At the receptor level, the compound binds both GIP and GLP-1 receptors and triggers downstream signalling that raises cyclic AMP in target cells. GLP-1 receptor activation is associated with glucose-dependent insulin release, slower gastric emptying, and reduced appetite signalling. GIP receptor activation contributes effects that are less completely characterised, and how much each receptor adds to the overall clinical response is still an open question. The two pathways appear to interact in a complementary rather than a purely additive way.
An extended fatty diacid moiety promotes binding to serum albumin, which slows renal clearance and extends the circulating half-life to roughly five days. That property supports once-weekly administration and largely explains the dosing interval described in clinical reports. Published data come mainly from large randomised programmes that evaluated glycaemic control and body weight over periods of many months. Long-term outcomes beyond those trial windows, including what happens after treatment stops, remain an active area of investigation.
Tirzepatide is a synthetic peptide built from thirty-nine amino acids. Its sequence is derived from native glucose-dependent insulinotropic polypeptide, or GIP, with several non-natural residues and a fatty diacid side chain attached through a linker. The molecule behaves as a dual agonist at two incretin receptors, GIP and GLP-1, instead of targeting a single receptor. This dual engagement separates it from earlier single-receptor incretin compounds and underpins most of its reported pharmacological activity.
Ornithology (from Ancient Greek ὄρνις (órnis) 'bird' and λόγος (lógos) 'study of') is a branch of zoology dedicated to the study of birds. Several aspects of ornithology differ from related disciplines, due partly to the high visibility and the aesthetic appeal of birds. It has also been an area with a large contribution made by amateurs in terms of time, resources, and financial support. Studies on birds have helped develop key concepts in biology including evolution, behaviour and ecology such as the definition of species, the process of speciation, instinct, learning, ecological niches, guilds, insular biogeography, phylogeography, and conservation. While early ornithology was principally concerned with descriptions and distributions of species, ornithologists today seek answers to very specific questions, often using birds as models to test hypotheses or predictions based on theories. Most modern biological theories apply across life forms, and the number of scientists who identify themselves as ornithologists has therefore declined. A wide range of tools and techniques are used in ornithology, both inside the laboratory and out in the field, and innovations are constantly made. Most biologists who recognise themselves as ornithologists study specific biology research areas, such as anatomy, physiology, taxonomy (phylogenetics), ecology, or behaviour.
== Biological function == Flavin reductases exist in a variety of organisms, including animals and bacteria. In luminous organisms, flavin reductase is important in the luciferase process. In an experiment with P. fischeri and B. harveyi cells, bioluminescence was increased as the in vivo concentration of flavin reductase was increased. This suggests a connection between either a flavin reductase-luciferase complex or reduced flavin and the luminescence process in bacteria. The bacteria oxidize the reduced flavin mononucleotide to oxidized FMN and transfer it through free fusion to generate light. In humans, flavin reductase often catalyzes an NADPH dependent reduction of flavin mononucleotide which occurs in methemoglobin in erythrocytes and the liver. It has also been suggested that flavin reductases play a role in the production of hydrogen peroxide. This would be biologically helpful as H2O2 assists the body in maintaining homeostatic microbiota. A study showed that women with lactobacillus that produced hydrogen peroxide were less likely to develop bacterial vaginosis prebirth. It was also seen in Trichomonas vaginalis that decreased levels of flavin reductase increased the cycling of metronidazole because flavin reductase has an antioxidative effect, which decreases oxygen levels, maintaining the metronidazole population.
==== Spirituality ==== While religion is often formalised and community-oriented, spirituality tends to be individually based and not as formalised. In a 2014 study, 320 children, ages 8–12, in both public and private schools, were given a Spiritual Well-Being Questionnaire assessing the correlation between spirituality and happiness. Spirituality – and not religious practices (praying, attending church services) – correlated positively with the child's happiness; the more spiritual the child was, the happier the child was. Spirituality accounted for about 3–26% of the variance in happiness. Meditation has been found to lead to high activity in the brain's left prefrontal cortex, which in turn has been found to correlate with happiness. A study using the Oxford happiness questionnaire on Brahma Kumaris Raja yoga meditators showed them having higher happiness than the control group. Yongey Mingyur Rinpoche has said that neuro scientists have found that with meditation, an individual's happiness baseline can change. Many people describe themselves as both religious and spiritual, but spirituality represents just one particular function of religion. Spirituality as related to positive psychology can be defined as "a search for the sacred". What is defined as sacred can be related to God, life itself, or almost any other facet of existence. It is viewed as having spiritual implications which are transcendent of the individual. Spiritual well-being addresses this human need for transcendence and involves social as well as existential well-being.
== Indications == D-dimer testing is of clinical use when there is a suspicion of deep venous thrombosis (DVTl), pulmonary embolism (PE) or disseminated intravascular coagulation (DIC). For DVT and PE, there are possible various scoring systems that are used to determine the a priori clinical probability of these diseases; the best-known is the Wells score.

Argon's complete octet of electrons indicates full s and p subshells. This full valence shell makes argon very stable and extremely resistant to bonding with other elements. Before 1962, argon and the other noble gases were considered to be chemically inert and unable to form compounds; however, compounds of the heavier noble gases have since been synthesized. The first argon compound with tungsten pentacarbonyl, W(CO)5Ar, was isolated in 1975. However, it was not widely recognised at that time. In August 2000, another argon compound, argon fluorohydride (HArF), was formed by researchers at the University of Helsinki, by shining ultraviolet light onto frozen argon containing a small amount of hydrogen fluoride with caesium iodide. This discovery caused the recognition that argon could form weakly bound compounds, even though it was not the first. It is stable up to 17 kelvins (−256 °C). The metastable ArCF2+2 dication, which is valence-isoelectronic with carbonyl fluoride and phosgene, was observed in 2010. Argon-36, in the form of argon hydride (argonium) ions, has been detected in interstellar medium associated with the Crab Nebula supernova; this was the first noble-gas molecule detected in outer space. Solid argon hydride (Ar(H2)2) has the same crystal structure as the MgZn2 Laves phase. It forms at pressures between 4.3 and 220 GPa, though Raman measurements suggest that the H2 molecules in Ar(H2)2 dissociate above 175 GPa.
In December 2019, the US Food and Drug Administration (FDA) announced that it learned that some metformin medicines manufactured outside the United States might contain a nitrosamine impurity called N-nitrosodimethylamine (NDMA), classified as a probable human carcinogen, at low levels. Health Canada announced that it was assessing NDMA levels in metformin. The European Medicines Agency provided an update on NDMA in metformin. In February 2020, the FDA found NDMA levels in some tested metformin samples that did not exceed the acceptable daily intake. In February 2020, Health Canada announced a recall of Apotex immediate-release metformin, followed in March by recalls of Ranbaxy metformin and in March by Jamp metformin. In May 2020, the FDA asked five companies to voluntarily recall their sustained-release metformin products. The five companies were not named, but they were revealed to be Amneal Pharmaceuticals, Actavis Pharma, Apotex Corp, Lupin Pharma, and Marksans Pharma Limited in a letter sent to Valisure, the pharmacy that had first alerted the FDA to this contaminant in metformin via a Citizen Petition. In June 2020, the FDA posted its laboratory results showing NDMA amounts in metformin products it tested. It found NDMA in certain lots of ER metformin and is recommending companies recall lots with levels of NDMA above the acceptable intake limit of 96 nanograms per day. The FDA is also collaborating with international regulators to share testing results for metformin.
(S)-Nicotine − this naturally occurring form of nicotine, found in tobacco plants at over 99% purity, is levorotatory with a specific rotation of [α]D(20°C)=–169.3°. (R)-Nicotine − this is the dextrorotatory form that is physiologically less active and less toxic than (S)-nicotine. The salts of (S)-nicotine are usually dextrorotatory; this conversion between levorotatory and dextrorotatory upon protonation is common among alkaloids. The hydrochloride and sulfate salts become optically inactive if heated in a closed vessel above 180 °C. The most common chemistry synthetic methods for generating nicotine yield a product that is approximately equal proportions of the S- and R-enantiomers. Tobacco-derived nicotine (>99% (S)-enantiomer) is distinguishable from synthetic nicotine (typically racemic, 50:50 (S)/(R)) by enantiomeric ratio analysis, although strategies exist for adjusting the relative levels of the enantiomers or performing a synthesis that only leads to the pure S-enantiomer. Synthetic stereospecific (S)-nicotine has become available on the market to consumers of electronic cigarette products. Nicotine enantiomers differ in their biological effects on animals.
