Identity and purity of tirzepatide are assessed mainly by reversed-phase high-performance liquid chromatography with ultraviolet detection, often paired with mass spectrometry. Because the molecule carries several modifications, gradient conditions are adjusted to resolve the intact peptide from deamidation and oxidation products. Enzymatic digestion followed by peptide mapping confirms the primary sequence and locates specific modifications. Quantitation in biological matrices typically uses liquid chromatography with tandem mass spectrometry after solid-phase extraction. Immunoassays are used less often, since antibody cross-reactivity with closely related peptides can bias results.
The peptide shares degradation routes common to modified peptides: deamidation of asparagine and glutamine residues, oxidation of methionine, and backbone hydrolysis under extreme pH. Lyophilized material is generally more stable than a solution, and residual water content directly affects the rate of hydrolysis. In liquid form, aggregation and visible particles can appear after agitation or repeated freeze-thaw cycles. Stability studies therefore track monomer content, aggregate content, and potency over months under defined temperature and humidity.
Long-term storage of the solid generally relies on temperatures at or below minus twenty degrees Celsius, while short-term working stocks may be held refrigerated. Light exposure is limited because photodegradation can alter side chains over extended periods. Solutions prepared for analysis are less stable than the dry powder and are typically used within the same working day. Buffer choice matters, since some aqueous conditions favor deamidation or oxidation at specific residues. Stability data are usually generated under defined accelerated conditions and then extrapolated with stated assumptions.
Identity and purity are established with reversed-phase high-performance liquid chromatography, often paired with mass spectrometry for confirmation of the expected mass. Peptide mapping after enzymatic digestion verifies the primary sequence and detects substitutions. Size-exclusion chromatography quantifies aggregates and fragments, which are the impurities most often tracked for peptides of this size. Residual solvents, counterions, and water content fall under separate tests described in pharmacopeial chapters. Circular dichroism or nuclear magnetic resonance may be used in research settings to probe secondary structure, though such methods are less common in routine release testing.
| Property | Value | Notes |
|---|---|---|
| Typical analytical method | Reversed-phase HPLC with UV detection | Often paired with mass spectrometry for identity |
| Common synonyms | GIP/GLP-1 dual agonist; LY3298176 | Development codes appear in earlier literature |
| Purity specification | Usually 95% or higher by HPLC area | Research-grade lots are often 98% or higher |
| Solution storage | 2–8 °C, protected from light | Short term; avoid repeated freeze-thaw cycles |
| Dry powder storage | −20 °C or below, desiccated | Protected from moisture and light |
Lyophilized material is generally held at -20 degrees Celsius or lower, desiccated and protected from light, where it remains stable for extended periods. Reconstituted or ready-to-use solution is usually kept at 2 to 8 degrees Celsius with minimal agitation. Repeated freeze-thaw cycles should be avoided because they promote aggregation and reduce the soluble monomer fraction. Shipment of frozen solid commonly uses dry ice, while refrigerated liquid moves with validated cold packs. Stability beyond documented periods is not established.
Characterization of the peptide relies on reversed-phase high-performance liquid chromatography for purity and related-substance profiling, with ultraviolet detection near 214 nanometers. Mass spectrometry confirms molecular mass and reveals modifications such as oxidation or deamidation. Peptide mapping after enzymatic digestion verifies the amino acid sequence, while amino acid analysis supplies compositional data. Circular dichroism and infrared spectroscopy are used to assess secondary structure, particularly the alpha-helical content that influences aggregation behavior in solution.
Common degradation routes include hydrolysis of labile amide bonds, deamidation of asparagine and glutamine residues, oxidation of methionine and tryptophan, and non-covalent aggregation. Aggregates can form during freeze-thaw cycling, at elevated pH, or when peptide concentration is high. Each route produces characteristic chromatographic or mass shifts that are tracked during stability studies. Whether a given minor impurity alters biological activity is often an open question, and specification limits are typically set on identity and purity rather than on functional data for trace species.
An extended fatty diacid moiety promotes binding to serum albumin, which slows renal clearance and extends the circulating half-life to roughly five days. That property supports once-weekly administration and largely explains the dosing interval described in clinical reports. Published data come mainly from large randomised programmes that evaluated glycaemic control and body weight over periods of many months. Long-term outcomes beyond those trial windows, including what happens after treatment stops, remain an active area of investigation.
Tirzepatide is a synthetic peptide built from thirty-nine amino acids. Its sequence is derived from native glucose-dependent insulinotropic polypeptide, or GIP, with several non-natural residues and a fatty diacid side chain attached through a linker. The molecule behaves as a dual agonist at two incretin receptors, GIP and GLP-1, instead of targeting a single receptor. This dual engagement separates it from earlier single-receptor incretin compounds and underpins most of its reported pharmacological activity.
=== Anti-aging and rejuvenation treatment === Solar UV radiation accelerates the skin ageing rate and contributes to abnormal cutaneous status by increasing matrix-metalloproteinase (MMP) expression in photo-damaged skin. MMPs consist of various proteinases that degrade collagen fibrils and other components of the cutaneous extracellular matrix. The commonly used therapies for skin ageing involve the use of retinoic acid, laser resurfacing, peels (trichloroacetic acid and CO2), injectable skin rejuvenation, and dermal fillers. In comparison, LEDT is a more non-ablative cutaneous rejuvenation approach with less safety concern and higher effectiveness. LECT with yellow LED (at a wavelength of 590 nm) contributes to the skin rejuvenation effects on nearly a thousand clinical samples. Researchers conclude that the use of red-light LEDT is an efficient collagen enhancement strategy. LEDT's combination therapy of different light wavelengths also shows greater efficacy. For instance, blue-light LEDT coupled with photosensitizers reveals better elasticity, less pigmentation, and more complexion of the skin.
== About == CoWIN serves the function of registration, appointment scheduling, identity verification, vaccination and certification of each vaccinated member. Registration for the vaccination slots can be booked on the same day or a few days prior. The platform has also been integrated in the Aarogya Setu and UMANG Apps. The certificate after COVID-19 vaccination can also be obtained through the platform. To expedite the development of this platform, several existing digital assets were leveraged, such as: Electronic Vaccine Intelligence Network (eVIN), Digital Infrastructure for Vaccination Open Credentialing (DIVOC), DigiLocker, Surveillance and Action for Events Following Vaccination (SAFE-VAC). CoWIN application was developed with five modules: the orchestration module; the vaccination cold chain module; the citizen registration module; the vaccinator module; and the certificate, feedback, and adverse event following immunization reporting module. As of now, eight vaccines can be registered on the platform in the country Covishield (18+), Covaxin (15+), Corbevax (12-14), Sputnik V (18+), Corbevax (12-14), Gemcovac (18+) Incovacc (18+), ZyCoV-D (18+) and Covovax (12+). In the future, the Health ministry is working on upgrading CoWIN for the effective implementation of India's universal immunization programme. Co-WIN platform will be used for booking slots for the routine vaccinations like Polio and Hepatitis.
== Etymology == The English word "tofu" comes from Japanese tōfu (豆腐, see below), which in turn borrows Chinese 豆腐 (Mandarin: dòufǔ or tòufu) 'bean ferment'. The earliest documentation of the word in English is in the 1704 translation of Domingo Fernández Navarrete's A Collection of Voyages and Travels, that describes how tofu was made. The word towfu also appears in a 1770 letter from the English merchant James Flint to Benjamin Franklin. The term "bean curd(s)" for tofu has been used in the United States since at least 1840.
=== Minor leagues (1949–51) === Mizell's first team was the Albany Cardinals of the Class D Georgia–Florida League. His first pitch for the team sailed 20 feet over the backstop. Overcoming homesickness, he went on to help Albany win the pennant. He had a 12–3 record and a 1.95 earned run average (ERA) for Albany, recording 175 strikeouts in 141 innings pitched. Promoted to the Class B Winston-Salem Cardinals of the Carolina League in 1950, Mizell got off to a 1–6 start. However, he went on to post a 17–7 record, finishing tied (with Wes Livengood) for fourth in the Carolina League in wins and second (behind Woody Rich) in ERA with a 2.48 mark. He struck out 227 batters in 207 innings, leading Winston-Salem to a championship. In the last game of the season, he hit a home run, the only professional one of his career, which he referred to as a $220 home run because fans passed a hat around and donated that sum in appreciation of his efforts. One night during the season, he sang country music over the public address system at South Side Park and rode around the field on a mule. In 1951, Mizell pitched for the Houston Buffaloes of the Class AA Texas League. The team held a "Vinegar Bend Night," flying in 32 visitors reportedly from the town in Alabama for the evening's game. Because the town was so small, Mizell said that some of the guests "must've come from the suburbs." He had 15 strikeouts in that game, a 3–1 loss to the Shreveport Sports. In another outing, against the Dallas Eagles, he struck out 18 batters, tying the league's single-game record.

=== Manuscript locations === Before his death, Tolkien negotiated the sale of the manuscripts, drafts, proofs and other materials related to his then-published works—including The Lord of the Rings, The Hobbit and Farmer Giles of Ham—to the Department of Special Collections and University Archives at Marquette University's John P. Raynor, S.J., Library in Milwaukee, Wisconsin, United States. After his death his estate donated the papers containing Tolkien's Silmarillion mythology and his academic work to the Bodleian Library at Oxford University. The Bodleian Library held an exhibition of his work in 2018, including more than 60 items which had never been seen in public before. In 2009 a partial draft of Language and Human Nature, which Tolkien had begun co-writing with Lewis but had never completed, was discovered at the Bodleian Library.
At 40 or more CAG repeats, full penetrance allele (FPA) exists. A "positive test" or "positive result" generally refers to this case. A positive result is not considered a diagnosis, since it may be obtained decades before the symptoms begin. However, a negative test means that the individual does not carry the expanded copy of the gene and will not develop HD. The test will tell a person who originally had a 50% chance of inheriting the disease if their risk goes up to 100% or is eliminated. Persons who test positive for the disease will develop HD sometime within their lifetimes, provided they live long enough for the symptoms to appear. At 36 to 39 repeats, incomplete or reduced penetrance allele (RPA) may cause symptoms, usually later in the adult life. The maximum risk is 60% that a person with an RPA will be symptomatic at age 65, and 70% at 75. At 27 to 35 repeats, intermediate allele (IA), or large normal allele, is not associated with symptomatic disease in the tested individual, but may expand upon further inheritance to give symptoms in offspring. With 26 or fewer repeats, the result is not associated with HD. Testing before the onset of symptoms is a life-changing event and a very personal decision. The main reason given for choosing to test for HD is to aid in career and family decisions. Predictive testing for Huntington's disease has been available via linkage analysis (which requires testing multiple family members) since 1986 and via direct mutation analysis since 1993.
== Treatment == Symptomatic treatment usually involves analgesics for both cranial and spinal CSF leaks. Initial measures can include rest, caffeine intake (via coffee or intravenous infusion), and hydration. Corticosteroids may provide transient relief for some patients. An abdominal binder, which increases intracranial pressure by compressing the abdomen, can temporarily relieve symptoms for some people. Sometimes a CSF leak will heal on its own. Otherwise, symptoms may last months or even years.
=== Discrepant reports on natural agency === In 2013, researchers Michel de Waard (then at Université Joseph Fourier, Grenoble and Grenoble Institute of Neuroscience, La Tronche) reported in Angewandte Chemie that tramadol was found in relatively high concentrations (>1%) in the roots of the African pin cushion tree, Nauclea latifolia, concluding that it was a natural product in addition to its being a later human synthetic, and presenting a putative biosynthetic hypothesis for its origin. In 2014, Michael Spiteller (Technische Universität Dortmund) and collaborators reported results, also in Angewandte Chemie, that supported the conclusion that the presence of tramadol in those tree roots was the result of tramadol having been ingested by humans and having been administered to cattle (by farmers in the region); Spiteller et al. presented data that tramadol and its metabolites were present in animal excreta, which they then argue contaminated soil around the trees. They further observed that tramadol and its mammalian metabolites were found in tree roots in the far north of Cameroon where the commercial drug was in use, but not in the south where it was not being administered. In 2016, Spiteller and colleagues followed up their preceding work with a radiocarbon analysis that supported their contention that the tramadol found in N. latifolia roots was of human synthetic origin rather being plant-derived.
A variety of other skeletal features are shared by dinosaurs. However, because they either are common to other groups of archosaurs or were not present in all early dinosaurs, these features are not synapomorphies. For example, as diapsids, dinosaurs ancestrally had two pairs of Infratemporal fenestrae (openings in the skull behind the eyes), and as members of the diapsid group Archosauria, had additional openings in the snout and lower jaw. Additionally, several characteristics once thought to be synapomorphies are now known to have appeared before dinosaurs, or were absent in the earliest dinosaurs and independently evolved by different dinosaur groups. These include an elongated scapula, or shoulder blade; a sacrum composed of three or more fused vertebrae (three are found in some other archosaurs, but only two are found in Herrerasaurus); and a perforate acetabulum, or hip socket, with a hole at the center of its inside surface (closed in Saturnalia tupiniquim, for example). Another difficulty of determining distinctly dinosaurian features is that early dinosaurs and other archosaurs from the Late Triassic epoch are often poorly known and were similar in many ways; these animals have sometimes been misidentified in the literature. Dinosaurs stand with their hind limbs erect in a manner similar to most modern mammals, but distinct from most other reptiles, whose limbs sprawl out to either side. This posture is due to the development of a laterally facing recess in the pelvis (usually an open socket) and a corresponding inwardly facing distinct head on the femur.

This differential equation is the classic equation of motion for charged particles. Together with the particle's initial conditions, it completely determines the particle's motion in space and time in terms of m/Q. Thus mass spectrometers could be thought of as "mass-to-charge spectrometers". When presenting data, it is common to use the (officially) dimensionless m/z, where z is the number of elementary charges (e) on the ion (z=Q/e). This quantity, although it is informally called the mass-to-charge ratio, more accurately speaking represents the ratio of the mass number and the charge number, z.
== History == Porous silicon was discovered by accident in 1956 by Arthur Uhlir Jr. and Ingeborg Uhlir at the Bell Labs in the U.S. At the time, the Uhlirs were in the process of developing a technique for polishing and shaping the surfaces of silicon and germanium. However, it was found that under several conditions a crude product in the form of thick black, red or brown film were formed on the surface of the material. At the time, the findings were not taken further and were only mentioned in Bell Lab's technical notes. In the early 1980s, researchers at the Royal Signals and Radar Establishment (RSRE) in Malvern, England, carried out the first systematic study of the formation and microstructure of porous silicon. In 1985, Beale, Benjamin, Uren, Chew and Cullis published two key papers establishing that porous silicon was not a deposited stain film, as previously assumed, but was formed by electrochemical etching of pores with aspect ratios exceeding 1000:1. Using cross-sectional transmission electron microscopy, they identified two distinct types of porous silicon microstructure, dependent on the dopant concentration: in heavily doped (degenerate) silicon, current transport through the Schottky barrier at the silicon–electrolyte interface proceeds by quantum mechanical tunnelling, while in lightly doped (non-degenerate) silicon it occurs via thermionic emission.
== Research and career == In 2007 Delibegovic returned to the United Kingdom, where she was awarded the Research Councils UK 5-year tenure track fellowship to investigate obesity and ageing at the University of Aberdeen. She was made Professor in Diabetes Physiology in 2015 at the age of 38. The field of her research has focussed on the PTP1B phosphatase, the molecular mechanisms that cause diabetes and what the relationship is between diabetes and Alzheimer's disease. She has demonstrated that PTP1B can be used for targeted treatments, reaching the cells of specific organs without causing any side effects. In 2017 Delibegovic demonstrated a novel pharmaceutical, Trodusquemine, that could be used to treat type 2 diabetes and breast cancer. She went on to show that a single dose of Trodusquemine, the PTP1B inhibitor, could be used to reverse the effects of atherosclerosis. During the COVID-19 pandemic, Delibegovic, in collaboration with an SME Vertebrate Antibodies Ltd and the NHS Grampian, obtained funding from the Scottish Government/ Chief Scientist office to develop a diagnostic test that could support mass screening for coronavirus disease. Her long-term aim was to use artificial intelligence to identify which parts of the severe acute respiratory syndrome coronavirus 2 activated the body's immune system. At the time, other coronavirus disease tests available in the United Kingdom would not support rapid deployment, and several were unreliable. In May 2020, the tests developed by Delibegovic and her team were in development.
In a 2016 New Internationalist interview, Galloway speculated that an incident of sexual abuse from a colonel, which he suffered when he was 12, caused a "lifelong fear of being gay and this led me into ostentatious, rapacious heterosexual promiscuity". According to Galloway, he grew a moustache at the age of 15, and refused to shave it off when his headmaster objected. He decided, at the age of 18, never to drink alcohol; the reason was originally derived from comments by his father, and he has described alcohol as having a "very deleterious effect on people".
== Usage and Applications == COBRA has been used extensively in many research-based applications such as screening for DNA methylation changes at gene promoters in cancer studies, detecting altered methylation patterns at imprinted genes, and characterizing methylation patterns in the genome during development in mammals. In medicine, COBRA has been used as a tool to help diagnose human disease involving aberrant DNA methylation. Researchers utilized COBRA in conjunction with denaturing high performance liquid chromatography in the diagnosis of the genetic imprinting disorder Russell-Silver syndrome where hypomethylation of the imprinted gene H19 is responsible for the disorder in up to 50% of patients.

Reversed-phase liquid chromatography with ultraviolet detection is the usual approach, frequently combined with mass spectrometry for identity. Purity is reported as the area percentage of the main peak. Related impurities eluting near the main peak are usually summed and reported separately.
Removing water slows hydrolysis and limits aggregation, so dry powder retains its quality attributes longer than a solution. Suppliers define a shelf life and retest date for the dried form at specified temperatures. Once dissolved, the practical working lifetime shortens considerably.
It generally lists appearance, identity by mass, purity by chromatography, water or residual solvent content, and the methods used. Storage recommendations and a retest date are commonly included. Values are reported against a supplier specification rather than a single universal standard.
Water promotes hydrolysis and deamidation, so removing it slows degradation during transport and storage. The dry solid is also less prone to microbial growth than a solution. Reconstitution is therefore performed close to the point of use.
