Tirzepatide is a synthetic peptide that activates both the glucose-dependent insulinotropic polypeptide (GIP) and glucagon-like peptide-1 (GLP-1) receptors. This dual agonist profile distinguishes it from earlier incretin-based compounds that act on a single receptor. The molecule was engineered from the native GIP sequence and carries several non-natural residues that slow enzymatic breakdown. Researchers designed it to combine the insulinotropic effects of GIP signaling with the appetite and gastric-emptying effects associated with GLP-1 activation.
Development of tirzepatide took place under a research program that sought to test whether simultaneous engagement of two incretin receptors would produce greater metabolic effects than single-receptor agonism. Clinical trials were organized into the SURPASS series for type 2 diabetes and the SURMOUNT series for obesity and weight management. Regulatory clearance for type 2 diabetes came in 2022 in the United States, followed by approval for chronic weight management in 2023. The trial programs reported reductions in glycated hemoglobin and body weight relative to comparators, though long-term cardiovascular and durability data continue to accumulate.
At the receptor level, the compound binds both GIP and GLP-1 receptors and triggers downstream signalling that raises cyclic AMP in target cells. GLP-1 receptor activation is associated with glucose-dependent insulin release, slower gastric emptying, and reduced appetite signalling. GIP receptor activation contributes effects that are less completely characterised, and how much each receptor adds to the overall clinical response is still an open question. The two pathways appear to interact in a complementary rather than a purely additive way.
An extended fatty diacid moiety promotes binding to serum albumin, which slows renal clearance and extends the circulating half-life to roughly five days. That property supports once-weekly administration and largely explains the dosing interval described in clinical reports. Published data come mainly from large randomised programmes that evaluated glycaemic control and body weight over periods of many months. Long-term outcomes beyond those trial windows, including what happens after treatment stops, remain an active area of investigation.

Research and analytical settings increasingly require documentation of peptide origin and chain of custody. Certificate of analysis documents typically report purity by chromatographic area, mass confirmation, appearance, and residual solvent or counterion content. Independent verification by an accredited laboratory is common when a material will be used in a regulated study. Open questions remain about how well compendial methods transfer between laboratories, and about which impurity thresholds are meaningful for materials not intended for clinical use.
Peptide-based pharmaceutical products such as tirzepatide require controlled temperature management to preserve structural integrity. Manufacturer labeling generally specifies refrigeration at 2 to 8 degrees Celsius before first use, with protection from light and freezing. Exposure to repeated temperature cycling can promote aggregation or deamidation, which alters the analytical profile even when the visible solution appears unchanged. Once a product is in use, the permitted storage window and temperature range are defined by the specific labeled presentation rather than by general peptide rules.
Identity and purity assessment of tirzepatide relies primarily on reversed-phase high-performance liquid chromatography coupled with ultraviolet detection. Mass spectrometry, often in electrospray ionization mode, confirms the molecular mass and detects sequence-related impurities. Peptide mapping after enzymatic digestion provides residue-level confirmation of the backbone. Each method addresses a different question: chromatography for purity and related substances, mass measurement for identity, and mapping for sequence fidelity. No single technique covers all three.
Like most synthetic peptides of this size, the material is commonly supplied as a lyophilised powder that appears white to off-white. It dissolves in aqueous buffers and in mixtures of water with a small proportion of organic solvent, though the fatty acid portion reduces solubility in pure water relative to short peptides. Hygroscopic behaviour is reported for many peptide powders, so weighing is usually performed quickly and under controlled humidity. Working solutions are typically prepared fresh and kept cold.
Long-term storage of lyophilised peptide powder is generally at minus twenty degrees Celsius or colder, with desiccant and protection from light. Short-term storage at two to eight degrees Celsius is common during active use. In solution, stability depends strongly on pH, concentration, and the presence of preservatives, and hydrolysis or aggregation can develop over weeks. Published stability data specific to this molecule are limited, so recommended conditions for research material are usually extrapolated from general peptide handling practice rather than from a dedicated study.

has no real roots. While all of the above inequalities are true because a, b, and c must follow the basic triangle inequality that the longest side is less than half the perimeter, the following relations hold for all positive a, b, and c:
Semi-closed rebreather technology is sometimes used in diver carried surface supplied gas extenders, mainly to reduce helium use. Some units also function as an emergency gas supply using on-board bailout cylinders: The US Navy MK29 rebreather can extend the duration of the Flyaway Mixed Gas System diving operations by five times while retaining the original mixed-gas storage footprint on the support ship. The Soviet IDA-72 semi-closed rebreather has a scrubber endurance of 4 hours on surface supply, and bailout endurance at 200m of 40 minutes on on-board gas. The US Navy Mark V Mod 1 heliox mixed gas helmet has a scrubber canister mounted on the back of the helmet and an inlet gas injection system which recirculates the breathing gas through the scrubber to remove carbon dioxide and thereby conserve helium. The injector nozzle would blow 11 times the volume of the injected gas through the scrubber.

Often, only between 10 and 15 mites are involved in an infection. Scabies most often spreads during a relatively long period of direct skin contact with an infected person (at least 10 minutes) such as that which may occur during sexual activity or living together. Spread of the disease may occur even if the person has not developed symptoms yet. Crowded living conditions, such as those found in child-care facilities, group homes, and prisons, increase the risk of spread. Areas with a lack of access to water also have higher disease rates. Crusted scabies is a more severe form of the disease, not essentially different but an infestation by huge numbers of mites that typically only affects those with a poor immune system; the number of mites also makes them much more contagious. In these cases, the spread of infection may occur during brief contact or by contaminated objects. The mite is tiny and at the limit of detection with the human eye. It is not readily obvious; factors that aid in detection are good lighting, magnification, and knowing what to look for. Diagnosis is based either on detecting the mite (confirmed scabies), detecting typical lesions in a typical distribution with typical histological features (clinical scabies), or detecting atypical lesions or atypical distribution of lesions with only some histological features present (suspected scabies). Several medications are available to treat those infected, including oral and topical ivermectin, permethrin, crotamiton, and lindane creams.
As a prototypical transaminase, AST relies on PLP (vitamin B6) as a cofactor to transfer the amino group from aspartate or glutamate to the corresponding ketoacid. In the process, the cofactor shuttles between PLP and the pyridoxamine phosphate (PMP) form. The amino group transfer catalyzed by this enzyme is crucial in both amino acid degradation and biosynthesis. In amino acid degradation, following the conversion of α-ketoglutarate to glutamate, glutamate subsequently undergoes oxidative deamination to form ammonium ions, which are excreted as urea. In the reverse reaction, aspartate may be synthesized from oxaloacetate, which is a key intermediate in the citric acid cycle. Two isoenzymes are present in a wide variety of eukaryotes. In humans:
In 2018, Bio-Rad Laboratories filed a lawsuit against 10x Genomics stating that their linked-read technology infringed on three patents which had been licensed from Bio-Rad at the University of Chicago. Bio-Rad was awarded a sum of $23,930,716 by a jury. The 10x Genomics filed a motion for judgement as a matter of law (JMOL) but were denied in 2019, and the court proceedings concluded in 2020. Following this lawsuit, 10x Genomics discontinued their linked-read assay. An exception was made for linked-read products which had already been sold by the company prior to the lawsuit, allowing 10x Genomics to continue to provide those researchers with services such as support and warranty maintenance for this technology.
As of today and for the next few hundred years or so, caesium-137 and strontium-90 continue to be the principal source of radiation in the zone of alienation around the Chernobyl nuclear power plant, and pose the greatest risk to health, owing to their approximately 30-year half-life and biological uptake. An estimated area of 12000 km² of Germany is contaminated with caesium-137 following the Chernobyl disaster in 1986 with surface activity of 20 to 37 kBq/m2. This corresponds to 1.1% of all caesium-137 released in Europe after the Chernobyl accident. In Scandinavia, some reindeer and sheep exceeded the Norwegian legal limit (3000 Bq/kg) 26 years after Chernobyl. The Chernobyl caesium-137 has now decayed by more than half, but could have been locally concentrated by much larger factors.
In physics, the first law of thermodynamics is an expression of the conservation of total energy of a system. The increase of the energy of a system is equal to the sum of work done on the system and the heat added to that system:

==== Antidepressants ==== Antidepressants are one of the common drugs for fibromyalgia. Antidepressants can improve the quality of life for fibromyalgia patients in the medium term. Duloxetine and milnacipran have good evidence of substantial pain relief, with no increased risk for serious adverse effects. A 2023 meta-analysis found that duloxetine improved fibromyalgia symptoms, regardless of the dosage. SSRIs may be also be used to treat depression in people diagnosed with fibromyalgia. While amitriptyline has been used as a first-line treatment, the quality of evidence to support this use is poor. Very weak evidence indicates that a very small number of people may benefit from treatment with the tetracyclic antidepressant mirtazapine, however, for most, the potential benefits are not great and the risk of adverse effects and potential harm outweighs any potential for benefit. Tentative evidence suggests that monoamine oxidase inhibitors (MAOIs) such as pirlindole and moclobemide are moderately effective for reducing pain. Very low-quality evidence suggests pirlindole as more effective at treating pain than moclobemide. Side effects of MAOIs may include nausea and vomiting. For sleep quality, there is no good evidence showing that amitriptyline, milnacipran or duloxetine give improvement.
=== Macronutrients === Supplementation of protein in the diet of healthy adults increases the size and strength of muscles during prolonged resistance exercise training (RET); protein intakes of greater than 1.62 grams per kilogram of body weight a day did not additionally increase fat–free mass (FFM), muscle size, or strength, in a non-energy restricted context. Older lifters may experience less of an effect from protein supplementation on resistance training. It is not known how much carbohydrate is necessary to maximize muscle hypertrophy. Strength adaptations may not be hindered by a low-carbohydrate diet. A light, balanced meal prior to the workout (usually one to two hours beforehand) ensures that adequate energy and amino acids are available for the intense bout of exercise. The type of nutrients consumed affects the response of the body, and nutrient timing whereby protein and carbohydrates are consumed prior to and after workout has a beneficial impact on muscle growth. Water is consumed throughout the course of the workout to prevent poor performance due to dehydration. A protein shake is often consumed immediately following the workout. However, the anabolic window is not particularly narrow and protein can also be consumed before or hours after the exercise with similar effects. Glucose (or another simple sugar) is often consumed as well since this quickly replenishes any glycogen lost during the exercise period.

It binds and activates both the GIP and GLP-1 receptors, making it a dual incretin receptor agonist. Single-receptor GLP-1 agonists act on one target only. The dual profile is the defining pharmacological feature of the molecule.
A fatty diacid side chain promotes reversible binding to serum albumin. This association slows renal clearance and protects the peptide from rapid enzymatic degradation. The result is a prolonged circulation time that supports weekly administration.
The first regulatory approval, for type 2 diabetes, was granted in the United States in 2022. An additional approval for chronic weight management followed in 2023. Availability and approved indications vary by country and are set by each national regulator.
It acts as a dual agonist at the GIP receptor and the GLP-1 receptor. This broader targeting profile distinguishes it from selective GLP-1 agonists, which engage only one receptor.
