The peptide activates two G protein-coupled receptors, GIPR and GLP-1R. Binding triggers adenylyl cyclase activity and raises intracellular cyclic AMP in pancreatic beta cells, which potentiates insulin release when glucose is elevated. Signaling in the central nervous system is associated with reduced appetite and lower energy intake, while effects on gastric emptying and glucagon secretion are also reported. Because activity at both receptors is retained, the pharmacological profile is often described as incretin-based rather than selective for a single receptor.
After subcutaneous injection, absorption is gradual, and peak plasma levels are generally reached within one to three days. Albumin binding extends the apparent half-life to roughly five days, which supports a weekly administration schedule. Metabolism proceeds mainly through proteolytic cleavage of the peptide backbone and beta-oxidation of the fatty acid chain, rather than through cytochrome P450 pathways. Eliminated fragments are largely recycled through general protein turnover, and excretion of intact drug in urine is minimal. These properties distinguish the molecule from short-acting incretin mimetics.
In clinical research, tirzepatide has been studied in randomized controlled trials for glycemic control and body weight reduction. These trials typically measure changes in hemoglobin A1c and body weight over periods of several months. The drug is administered by subcutaneous injection, and its pharmacokinetic profile supports once-weekly dosing. Post-marketing surveillance continues to evaluate long-term outcomes and rare adverse events.
Tirzepatide is a synthetic peptide that acts as a dual agonist at the glucose-dependent insulinotropic polypeptide (GIP) and glucagon-like peptide-1 (GLP-1) receptors. The molecule contains 39 amino acids and features a C20 fatty diacid moiety attached via a linker, which promotes albumin binding and extends its circulating half-life. Its sequence incorporates non-natural amino acids and modifications that reduce susceptibility to degradation by dipeptidyl peptidase-4. This dual receptor activity distinguishes it from selective GLP-1 receptor agonists.

Development of tirzepatide took place under a research program that sought to test whether simultaneous engagement of two incretin receptors would produce greater metabolic effects than single-receptor agonism. Clinical trials were organized into the SURPASS series for type 2 diabetes and the SURMOUNT series for obesity and weight management. Regulatory clearance for type 2 diabetes came in 2022 in the United States, followed by approval for chronic weight management in 2023. The trial programs reported reductions in glycated hemoglobin and body weight relative to comparators, though long-term cardiovascular and durability data continue to accumulate.
The peptide backbone contains 39 amino acids and includes alpha-aminoisobutyric acid residues, which are not among the standard proteinogenic set. A C20 fatty diacid moiety is attached through a linker, allowing the compound to bind serum albumin and extend its circulation time. This albumin binding is the main reason the molecule supports once-weekly administration rather than more frequent dosing. The measured molecular mass is approximately 4,813 daltons, placing it firmly in the peptide rather than small-molecule class.
Tirzepatide is a synthetic peptide that activates both the glucose-dependent insulinotropic polypeptide (GIP) and glucagon-like peptide-1 (GLP-1) receptors. This dual agonist profile distinguishes it from earlier incretin-based compounds that act on a single receptor. The molecule was engineered from the native GIP sequence and carries several non-natural residues that slow enzymatic breakdown. Researchers designed it to combine the insulinotropic effects of GIP signaling with the appetite and gastric-emptying effects associated with GLP-1 activation.
Tirzepatide is a synthetic peptide of 39 amino acids that carries a C20 fatty diacid side chain attached through a linker. Its molecular formula is C225H348N48O68, and its molecular weight is about 4813 daltons. The compound belongs to the incretin mimetic class and is administered by subcutaneous injection. The fatty acid chain promotes binding to serum albumin, which slows renal clearance and extends the circulation time of the molecule. It was identified during screening of sequences derived from glucose-dependent insulinotropic polypeptide.
Tirzepatide activates both the glucose-dependent insulinotropic polypeptide receptor and the glucagon-like peptide-1 receptor, making it a dual agonist rather than a selective agent. Engagement of the GLP-1 receptor is linked to glucose-dependent insulin release, slower gastric emptying, and reduced appetite signalling. The relative contribution of the GIP arm remains an active research question; proposed roles include improved insulin sensitivity and altered adipose tissue handling. Receptor occupancy studies suggest the molecule interacts with both targets at circulating concentrations achieved during therapy.
== Oxygen-18 == Oxygen-18 (18O, Ω) is one of the stable isotopes of oxygen, with roughly 0.20% abundance, and considered one of the environmental isotopes. Most 18O is produced when 14N (made abundant from CNO burning) captures a 4He nucleus, becoming 18F. This quickly (half-life around 110 minutes) beta decays to 18O making that isotope common in the helium-rich zones of stars. Temperatures on the order of 109 kelvins are needed to fuse oxygen into sulfur. Fluorine-18 is usually produced by irradiation of 18O-enriched water with high-energy (about 18 MeV) protons prepared in a cyclotron or a linear accelerator, yielding an aqueous solution containing 18F as fluoride ion. This solution is then used for rapid synthesis of a labeled molecule, often with the fluorine atom replacing a hydroxy group. The labeled molecules or radiopharmaceuticals have to be synthesized after the radiofluorine is prepared, as the high energy proton radiation would destroy the molecules. Large amounts of oxygen-18 enriched water are used in positron emission tomography centers, for on-site production of 18F-labeled fluorodeoxyglucose (FDG). Measurements of the 18O/16O ratio (known as δ18O) are often used in paleoclimatology. Water molecules with a lighter isotope are slightly more likely to evaporate and less likely to fall as precipitation, so Earth's freshwater and polar ice have slightly less (0.1981%) 18O than air (0.204%) or seawater (0.1995%). This disparity allows the study of historical temperature patterns via the analysis of ice cores.
== History == Hans Christian Hagedorn (1888–1971) and August Krogh (1874–1949) obtained the rights for insulin from Frederick Banting and Charles Best in Toronto, Canada. In 1923 they formed Nordisk Insulin laboratorium, and in 1926 with August Kongsted he obtained a Danish royal charter as a non-profit foundation. In 1936, Hagedorn and B. Norman Jensen discovered that the effects of injected insulin could be prolonged by the addition of protamine obtained from the "milt" or semen of river trout. The insulin would be added to the protamine, but the solution would have to be brought to pH 7 for injection. University of Toronto, Canada later licensed protamine zinc insulin (PZI), to several manufacturers. This mixture only needs to be shaken before injection. The effects of PZI lasted for 24–36 h.
=== Pattern-based measures === Earlier methods for measuring convergence incorporate ratios of phenotypic and phylogenetic distance by simulating evolution with a Brownian motion model of trait evolution along a phylogeny. More recent methods also quantify the strength of convergence. One drawback to keep in mind is that these methods can confuse long-term stasis with convergence due to phenotypic similarities. Stasis occurs when there is little evolutionary change among taxa. Distance-based measures assess the degree of similarity between lineages over time. Frequency-based measures assess the number of lineages that have evolved in a particular trait space.
Given the overwhelming Allied air superiority, the Luftwaffe put its effort into the development of aircraft of such high performance that they could operate with impunity, but which also made bomber attack much more difficult, merely from the flight velocities they achieved. Foremost among these were the Messerschmitt Me 163B point-defense rocket interceptors, which started their operations with JG 400 near the end of July 1944, and the longer-endurance Messerschmitt Me 262A jet fighter, first flying with the Gruppe-strength Kommando Nowotny unit by the end of September 1944. In action, the Me 163 proved to be more dangerous to the Luftwaffe than to the Allies and was never a serious threat. The Me 262A was a serious threat, but attacks on their airfields neutralized them. The pioneering Junkers Jumo 004 axial-flow jet engines of the Me 262As needed careful nursing by their pilots, and these aircraft were particularly vulnerable during takeoff and landing. Lt. Chuck Yeager of the 357th Fighter Group was one of the first American pilots to shoot down an Me 262, which he caught during its landing approach. On 7 October 1944, Lt. Urban L. Drew of the 361st Fighter Group shot down two Me 262s that were taking off, while on the same day, Lt. Col. Hubert Zemke, who had transferred to the Mustang-equipped 479th Fighter Group, shot down what he thought was a Bf 109, only to have his gun camera film reveal that it may have been an Me 262. On 25 February 1945, Mustangs of the 55th Fighter Group surprised an entire Staffel of Me 262As at takeoff and destroyed six jets.
=== Minipreparation === Minipreparation of plasmid DNA is a rapid, small-scale isolation of plasmid DNA from bacteria. Commonly used miniprep methods include alkaline lysis and spin-column based kits. It is based on the alkaline lysis method. The extracted plasmid DNA resulting from performing a miniprep is itself often called a "miniprep". Minipreps are used in the process of molecular cloning to analyze bacterial clones. A typical plasmid DNA yield of a miniprep is 5 to 50 μg depending on the cell strain. Miniprep of a large number of plasmids can also be done conveniently on filter paper by lysing the cell and eluting the plasmid on to filter paper.

=== Plasma cortisol concentration === Resting plasma cortisol may be slightly elevated in affected horses, but is commonly within normal limits or below normal. Additionally, elevations may occur secondary to stress, concurrent disease, and due to individual variation. Therefore, resting cortisol levels alone are not adequate to diagnose or rule out the presence of PPID.
=== Combination therapy === Combinations of finasteride, minoxidil and ketoconazole are more effective than individual use. Combination therapy of LLLT or microneedling with finasteride or minoxidil demonstrated substantive increases in hair count.
Static self-assembly is significantly slower compared to dynamic self-assembly as it depends on the random chemical interactions between particles. Self assembly can be directed in two ways. The first is by manipulating the intrinsic properties which includes changing the directionality of interactions or changing particle shapes. The second is through external manipulation by applying and combining the effects of several kinds of fields to manipulate the building blocks into doing what is intended. To do so correctly, a high level of direction and control is required and developing a simple, efficient method to organize molecules and molecular clusters into precise, predetermined structures is crucial.
Cod is popular as a food with a mild flavour and a dense, flaky white flesh. Cod livers are processed to make cod liver oil, an important source of vitamin A, vitamin D, vitamin E and omega-3 fatty acids (EPA and DHA).
== Development == The mod was announced on March 6, 2014, by Coburn on Mod DB, citing the original Half-Life game and user-created mods as a reason to "give back to the community." Coburn stated that the mod was his "first real modding effort," creating a small campaign before expanding it during his spare time. The development took five years to complete before releasing on August 10. During the development, his child was born with a heart complication and required surgery at the Royal Hospital for Children, Glasgow and recovered. After the mod was complete, he requested all fans who wanted to donate to the hospital. The mod uses voice lines from the original Half-Life, as well as Half-Life: Opposing Force, Half-Life: Blue Shift, Half-Life: Decay, and Half-Life 2: Episode Two. On August 12, 2021, a team of developers announced a remake in Black Mesa called Black Mesa: Candidate Twelve after half a year in production. However this was confirmed to be on a hiatus in 2025 as they moved onto other projects.

It is a synthetic linear peptide that acts as a dual agonist at the GIP and GLP-1 receptors. It combines a modified incretin backbone with a fatty diacid side chain that extends its circulation time. It is not a small-molecule drug and is not orally absorbed in its native form.
Single-receptor agents act predominantly at GLP-1R. Tirzepatide retains activity at GIPR, which is expressed in pancreatic islets, adipose tissue, and the central nervous system. The contribution of each receptor to the overall response is still an area of active study, and the relative weight of GIPR signaling in humans is not fully settled.
The fatty diacid moiety binds serum albumin, which reduces renal filtration of the active peptide. Additional resistance to enzymatic breakdown comes from non-natural residues in the backbone. Together these features produce an apparent half-life of about five days and permit weekly dosing intervals.
It activates both GIP and GLP-1 receptors. This dual action differentiates it from selective GLP-1 agonists.
